Set Appliance State:

Set Multimeter to AC Volts 
DHW Demand
Place a link between the Common and Calling connections of the cylinder thermostat
Ensure the link is safe and secure then refit the thermostats cover
Ensure the link is safe and secure then refit the thermostats cover

Measurement:
Once the link is in place the cylinder thermostat is bypassed and
the hot water should now fire regardless of the cylinder temperature.
Measure the voltage on the Brown wire to the Hot Water zone valve.
1: Is mains voltage present?
2: If voltage is now present on the brown wire, does the valve move to the open position?
Once test is completed remove the link.
Note: The colour and position of the wires will vary between systems and components.
