Set Appliance State:
Mains Power Off
Set Multimeter to AC Volts 
Turn Hot Water Tap On
Selector Switch in Hot Water Position

Measurement:
Once the boiler begins its ignition sequence (pump starts to run), monitor the voltage between X8 (flame sense lead) at the PCB and a suitable earth point.
Voltage should be approximately 140V AC.
Once the burner ignites the voltage should drop to 135V AC or lower.
Note:
Some voltage may be present with no demand however this is not relevant to the test.
